Common Side Effects

Pralatrexate has been studied extensively in clinical trials. In these studies, the side effects that occurred in a group of people using the drug were carefully documented. As a result, it was possible to see what problems occurred and how often they appeared.
 
In clinical studies, the most common pralatrexate side effects included:
 
  • Sores and swelling in the mouth -- in up to 70 percent of people
  • Low platelet counts (which could cause abnormal bruising and bleeding) -- up to 41 percent
  • Nausea -- up to 40 percent
  • Fatigue -- up to 36 percent
  • Anemia (low red blood cell counts) -- up to 34 percent
  • Constipation -- up to 33 percent
  • Fever -- up to 32 percent
  • Swelling of the arms, hands, legs, ankles, or feet -- up to 30 percent
  • Cough -- up to 28 percent
  • Nosebleeds -- up to 26 percent.
 
Other common reactions, occurring in 10 percent to 25 percent of people, included:
 
  • Vomiting
  • Decreased white blood cells (which could increase the risk for infection)
  • Diarrhea
  • Shortness of breath
  • Decreased appetite
  • Low levels of potassium in the blood (hypokalemia)
  • Rash
  • Itching
  • Sore throat
  • Elevated liver enzymes
  • Abdominal (stomach) pain
  • Arm or leg pain
  • Night sweats
  • Weakness or lack of energy
  • Increased heart rate
  • Upper respiratory tract infection, such as the common cold.
 

Final Thoughts

You may experience some or none of the side effects listed in this article. Unfortunately, there is no way for your healthcare provider to know beforehand if you will have problems with a medicine that you have never tried.
 
Therefore, make sure to let your healthcare provider know if you develop any side effects while receiving pralatrexate or if something "just does not seem right." While it may not be related to the medication, your healthcare provider will be able to diagnose and treat the problem.
